GLaDOS is the "bad guy" in the Portal video game. The player has to do a bunch of tests, at the direction of GladOS, a computer, that entices/encourages the player with the promise of cake. Throughout the game there are hints that the promise of cake is not going to be fulfilled/true. And thus "the cake is a lie".

To explain the name: GLaDOS is a robot in the game Portal, the premise of which is you are a test subject in Aperture Labs. You realize over the course of the game that GLaDOS is not an ally, often via dark humor. There is a promise of cake is an incentive to compete the tests. By game's end you realize "the cake is a lie" and that GLaDOS is AI gone rogue.
